i don't really understand the sirtuin research. David Sinclair makes a big deal about the importance of the sirtuins, but overexpression of them doesn't consistently extend lifespan or improve health. resveratrol has failed to live up to the hype. part of that might have to do with poor bioavailability. i agree with him that epigenetic dysregulation is the primary cause of aging, but that doesn't mean a sirtuin activator will do much.
the case for NMN or NR appears stronger, but it might not be related to the sirtuins. NAD is important in a variety of cellular functions.
rapamycin is easily the most promising anti-aging drug available.

Well the NMN and NR boosts NAD which seems to extend lifespan because they activate sirtuins.
Rapamycin I have heard is dangerous because it is an immunosuppressant and they side effects may make it less appealing to use or cancel out its observed benefits.
David Sinclair takes Metformin, which he talks up more because it works on universal aging pathways, but the idea of taking a diabetes drug for aging strikes me as strange.
