I've heard the aging scientists say once people are past 80, the level of damage would be difficult to reverse. If you are around 60ish or younger when this becomes viable, you will stand to benefit. They believe the first person who will live to be 1,000 years old is already alive.

I think they will throw untold resources at this because they believe that they'll get it to work in time. For the youngest boomers, it might. But once you're past a certain age it's tough to reverse the damage. Though so much of this is still hypothetical, it's difficult to say.

Eventually when they die, it will be up to us to take the rotted remains of society and the decrepit infrastructure Boomery has built and turn it into something better, like Great Renewal.

At some point, all forms of damage will be reversible. Treatments like senolytics and epigenetic reprogramming will have probably only work well if there has been limited tissue remodeling though. I think that's a good reason to take anti-aging drugs, even if you are young. Hopefully boomers are fucked.

i don't really understand the sirtuin research. David Sinclair makes a big deal about the importance of the sirtuins, but overexpression of them doesn't consistently extend lifespan or improve health. resveratrol has failed to live up to the hype. part of that might have to do with poor bioavailability. i agree with him that epigenetic dysregulation is the primary cause of aging, but that doesn't mean a sirtuin activator will do much.

the case for NMN or NR appears stronger, but it might not be related to the sirtuins. NAD is important in a variety of cellular functions.

rapamycin is easily the most promising anti-aging drug available.

Well the NMN and NR boosts NAD which seems to extend lifespan because they activate sirtuins.

Rapamycin I have heard is dangerous because it is an immunosuppressant and they side effects may make it less appealing to use or cancel out its observed benefits.

David Sinclair takes Metformin, which he talks up more because it works on universal aging pathways, but the idea of taking a diabetes drug for aging strikes me as strange.

It is not at all clear NR or NMN lifespan extension is mediated through increased sirtuin activity. It could be a number of different things causing it that are NAD dependent.

The immunosuppressive stuff is overblown. They tested it in elderly people and saw improved immune function. If you are taking a dose that would be normally prescribed to a transplant patient you will have problems. Smaller dosages can suppress SASP.
